Blahodárné účinky střídmé konzumace vína souvisejí s obsahem a složením polyfenolic-kých látek. Jejich antioxidační a protinádorové účinky byly mnohokrát prokázány v růz-ných studiích. Cílem diplomové práce bylo sledovat vlivy polyfenolů extrahovaných z révy vinné na proliferační aktivitu nádorových a imortalizovaných buněk. Antiproliferační efekt byl hodnocen in vitro za použití různých koncentrací polyfenolických extraktů (1, 5, 10, 25 a 50 ?g/ml). Studovány byly tři odrůdy révy vinné (Burgunda modrá, Frankovka a Morav-ský muškát) a tři typy buněčných linií (HaCaT, HepG2, NIH/3T3). Proliferace buněčných linií byla vyhodnocena pomocí MTT testu a detekce apoptózy byla provedena za použití průtokové cytometrie. |

Beneficial effects of moderate wine consumption are related to the content and compositi-on of polyphenolic substances. Their antioxidant and anticancer effects have been demon-strated in various studies for many times. The aim of this thesis was to determine the ef-fects of polyphenols extracted from the grapes on the proliferative activity of tumor and immortalized cells. Antiproliferative effect was evaluated in vitro using different concent-rations of polyphenolic extracts (1, 5, 10, 25 a 50 ?g/ml). The three grapes varieties (Blue Burgundy, Lemberger and Moravian Muscat) and three types of cell lines (HaCaT, HepG2, NIH/3T3) were studied. The cell proliferation was evaluated using the MTT assay and de-tection of apoptosis was performed using flow cytometry. |
